Yes, it is possible to block .exe's from being downloaded in your network. Doing so is just a matter of preventing your clients from accessing URL's that end with .exe. This is easy to do using the WWW Proxy Policies.
I have included basic instructions below. These instructions assume that you have not already made changes to the WWW Proxy Policies.
1. Gatekeeper -> Services -> WWW Proxy Server -> Policies
2. Remove any policies that already exisiting them by selecting them and pressing the remove button
3. Click the Add button to make a new policy
4. Goto the Advanced tab
5. Select "Specify which request this recipient has rights for"
6. Click Add Filter
7. Click Add Crtierion
8. Select "This criterion is NOT met if - HTTP URL - ends with - .exe"
9. Click OK, then Apply to finalise the change
